My hope is that is keeps going back in time, so maybe you start as a young Dutch or Hosea. Or one of the members that dies, so you continue on as a different gang member.
Protagonist? Definitely the Pinkerton's.

Nah give us the Jack Marston revenge arc where he hunts down all the men who killed his dad, add in some background about what happened after his death and boom it may be good I'd play it.

The problem with that is, in going further into the future, you are going to be driving cars and bootlegging booze. Moves it too far away from the wild west.

Doubt it. They first have to release GTA VI, then they might start working on it. They might not even work on it, and instead choose to work on GTA VII depending on the success of GTA VI.
