Im confused as to the difficulty youre having?

to find background spots you may gave missed
create a new layer and fill it with a colour that will bring it out and show you
remove those background pieces then save as a png.

op your not saving as a gif are you? because gifs have some transparancy issues, if you want a clean transparancy then you need to save as png o-o
